Geminiviridae), к которым и относится вирус желтой курчавости листьев томата (TYLCV).В связи с тем, что TYLCV имеет фитосанитарный статус и регулируется в соответствии с Решением Совета Евразийской экономической комиссии от 30 ноября 2016 г. № 158, особенно важно обеспечить его раннее выявление и идентификацию. В рамках настоящего исследования было принято решение определить возможность выделения ДНК TYLCV из особей табачной белокрылки, хранящихся разными способами – в 70%-м растворе этилового спирта и в замороженном состоянии. Питание данных особей происходило на растениях томата, зараженных TYLCV. Сравнивали выделение ДНК разными коммерческими наборами, после чего проводили измерение концентрации ДНК. Затем осуществляли постановку ПЦР в режиме реального времени.Дополнительно с целью анализа генетической последовательности участка гена цитохромоксидазы B. tabaci после выделения тотальной НК ставили классическую ПЦР к искомому фрагменту. Как показало исследование, ДНК-продукт был детектирован при обоих способах выделения, указанные методы применимы на практике. Так как в случае с образцами табачной белокрылки, хранящимися в 70%-м растворе этанола, наблюдали ингибирование реакции при выделении набором «Проба-НК», к использованию рекомендуется набор «ФитоСорб».</p></abstract><trans-abstract xml:lang="en"><p>Silverleaf whitefly (Bemisia tabaci Genn.) is one of the key vectors of phytopathogenic viruses, primarily begomoviruses (family Geminiviridae), which include tomato yellow leaf curl virus (TYLCV).Since TYLCV has phytosanitary status and is regulated in accordance with Decision No. 158 of the Council of the Eurasian Economic Commission dated November 30, 2016, it is particularly important to ensure its early detection and identification. This study aimed to determine the feasibility of isolating TYLCV DNA from silverleaf whitefly individuals stored in different ways — in a 70% ethanol solution and frozen. These individuals fed on TYLCV-infected tomato plants. DNA extraction using different commercial kits was compared, after which DNA concentrations were measured. Real-time PCR was then performed.Additionally, to analyze the genetic sequence of the B. tabaci cytochrome oxidase gene fragment after total NC isolation, classical PCR was applied to the target fragment. The study demonstrated that the DNA product was detected using both isolation methods, and these methods are applicable in practice.
