Диагностика осложнена тем, что заболе­вание вызывается четырьмя видами бактерий рода Xanthomonas. Основным путем распространения бактериоза на дальние расстояния являются семе­на. Европейская и Средиземноморская организа­ция по карантину и защите растений (ЕОКЗР) включила группу возбудителей черной бактериальной пятнистости в Список А2 перечня карантинных объектов. Выявление и идентификация фитопато­генов на всех этапах производства томата и перца позволят вовремя принять меры по защите расте­ний, что в значительной степени снизит потери урожая и повысит экономическую эффективность производства отечественных овощей. Существую­щие схемы диагностики болезни подразумевают большие трудозатраты и длительный срок прове­дения исследований, что неприемлемо в рутинной лабораторной диагностике. В статье представле­ны результаты исследований диагностики одного из возбудителей черной бактериальной пятнисто­сти (X. euvesicatoria pv. euvesicatoria). Для установ­ления степени влияния растительной матрицы на конечные результаты исследований образцов нами был проведен ряд опытов по искусственному заражению X. euvesicatoria pv. euvesicatoria экстрактов вегетативных частей и семян томата. В ходе испытания методики подготовки проб с дальнейшим выделением ДНК и ПЦР-анализом искусственно зара­женных образцов установлено, что ингибирования ПЦР не происходит. Аналитическая чувствитель­ность тест-системы для диагностики X. euvesicatoria pv. euvesicatoria производства ООО «АгроДиагности­ка» составила 103 КОЕ/мл. Исследования проведены на базе ФГБУ «ВНИИКР».</p></abstract><trans-abstract xml:lang="en"><p>Bacterial spot of tomato is worldwide spread and causes significant economic damage in tomato and pepper cultivation. Diagnosis is complicated by the fact that the disease is caused by four types of bacteria of the genus Xanthomonas. Seeds are the main pathway for bacteriosis over long distances. The European and Mediterranean Plant Protection Organization (EPPO) has included a group of causative agents of black bacterial spot in List A2 of the List of Quarantine Objects. Detection and identification of phytopathogens at all stages of tomato and pepper cultivation will allow taking timely measures to protect plants, which will significantly reduce crop losses and increase the economic efficiency of domestic vegetable production. Existing schemes for diagnosing the disease imply large labor costs and a long period of research, which is unacceptable in routine laboratory diagnosis. The article presents the results of studies on the diagnosis of one of the pathogens of black bacterial spot (X. euvesicatoria pv. euvesicatoria). To establish the degree of influence of the plant matrix on the final results of the samples study, we conducted a series of experiments on artificial infection with X. euvesicatoria pv. euvesicatoria of extracts of tomato vegetative parts and seeds. In the course of testing the sample preparation technique with further DNA isolation and PCR analysis of artificially infected samples, it was found that PCR inhibition does not occur. Analytical sensitivity of the diagnostic test system for X. euvesicatoria pv. euvesicatoria produced by AgroDiagnostics was 103 cfu/ml.
